چکیده
The major seed storage protein of rice (Oryza sativa L.), glutelin, is encoded by a smal1 multigene family of about 10 copies per haploid genome. Eight genes have a)ready been sequenced at both the cDNA and genomic DNA levels (Takaiwa et al. 1987a, b, 1989, 1991a, c, Okita et al, 1989), All the glutelin genes are specifically expressed in the maturing seed (Okita et al. 1989, Takaiwa et al. 1991a, b), Although it is considered that these genes are controlled by common regulatory mechanisms, there is little sequence homology among the genes other than a few conserved sequences, such as the AACA (AACAAACTCTATC), GCN4 (ATGAGICTCAT) and GCAA (Gl CCAAAATGA) motifs (Takaiwa et al. 1991c, 1996). Characterization of a new rice glutelin gene GluD-1 expressed in the starchy endosperm
A new glutelin gene, designated GluD-1, has been discovered by comparing the seed storage proteins from 48 japonica and indica rice cultivars on SDS-PAGE gels. Evidence that GluD-1 is a member of the glutelin family was provided by Western blots using anti-glutelin antiserum and by mapping the gene to the chromosomal glutelin gene cluster. The limited GluD-1 size polymorphism among the rice var...
متن کاملSmall GTPase Sar1 is crucial for proglutelin and α-globulin export from the endoplasmic reticulum in rice endosperm
Rice seed storage proteins glutelin and α-globulin are synthesized in the endoplasmic reticulum (ER) and deposited in protein storage vacuoles (PSVs). Sar1, a small GTPase, acts as a molecular switch to regulate the assembly of coat protein complex II, which exports secretory protein from the ER to the Golgi apparatus. To reveal the route by which glutelin and α-globulin exit the ER, four putat...
متن کاملLow glutelin content1: a dominant mutation that suppresses the glutelin multigene family via RNA silencing in rice.
Low glutelin content1 (Lgc1) is a dominant mutation that reduces glutelin content in rice grains. Glutelin is a major seed storage protein encoded by a multigene family. RNA gel blot and reverse transcriptase-mediated PCR analyses revealed that Lgc1 acts at the mRNA level in a similarity-dependent manner. In Lgc1 homozygotes, there is a 3.5-kb deletion between two highly similar glutelin genes ...
متن کاملProteomic Analysis Reveals Proteins Involved in Seed Imbibition under Salt Stress in Rice
Enhancement of salinity tolerance during seed germination is very important for direct seeding in rice. In this study, the salt-tolerant japonica landrace Jiucaiqing was used to determine the regulators that are involved in seed imbibition under salt stress. Briefly, the comparative proteomic analysis was conducted between dry (0 h) and imbibed (24 h) seeds with 150 mM NaCl. Under salt stress, ...
متن کاملLack of Globulin Synthesis during Seed Development Alters Accumulation of Seed Storage Proteins in Rice
The major seed storage proteins (SSPs) in rice seeds have been classified into three types, glutelins, prolamins, and globulin, and the proportion of each SSP varies. It has been shown in rice mutants that when either glutelins or prolamins are defective, the expression of another type of SSP is promoted to counterbalance the deficit. However, we observed reduced abundances of glutelins and pro...
